Lab research areas

1. Molecular genetics and cellular mechanisms of neurodegeneration in Parkinson’s disease and related neurodegenerative disorders

2. Development of cell-based assays to screen small molecules, drugs and natural compounds with the potential to block neuronal loss

3. Importance of PINK1-regulated mitochondrial quality control and mitochondrial dynamics in neurodegeneration, neurogenesis and tumorigenesis

4. Viral gene transfer (AAV, retroviruses, lentiviruses) and CRISPR/Cas9 gene editing for direct in vivo modeling and gene therapy of neurodegenerative disorders (Parkinson’s, Huntington’s, amyotrophic lateral sclerosis)

 

Lab techniques, models and tools

1. Generation of knockout/transgenic mice

2. Wide range of molecular biology, cell biology and biochemistry methods

3. Culture and genetic modification of primary cells (MEFs, astrocytes, neurons, stem cells)

4. Isolation of hippocampal neural stem cells, characterization of adult neurogenesis in the hippocampus of mice

5. Tissue sectioning, immunohistochemistry, histology

6. 3D image generation and analysis (Vaa3D)

7. Production and purification of recombinant AAV, retroviruses and lentiviruses, viral gene transfer to the brain and primary cells

8. Stereotaxic surgery

9. Light, confocal and electron microscopy

10. Isolation and purification of mitochondria

11. Real-time measurements of mitochondrial respiration and glycolysis in living cells and with isolated mitochondria (Seahorse Flux Analyzer)

12. Mouse behavior (depression, anxiety, motor function)
